Does anyone know of a website similar to MapQuest where I could request a cycling route between locations and be shown a map, or is there perhaps a website with a map of cycle paths in Cologne from which I could work out the best way to get from A to B?

I don't know any websites but a good bookshop should have a local cyclists map you can buy.

Does GoogleEarth show cycle routes? Someday perhaps.
If you've been spoilt by the Ordnance Survey maps you get in the UK (do you have these in Oz too?), then you'll be disappointed to learn that there's no nationwide equivalent here. The closest you can get - at least in NRW - is the series of maps from the Landesvermessungsamt.
They do both Freizeitkarten (1:50000) with foot- and cycle paths covering the whole of NRW, and Wanderkarten (1:25000) for areas of special interest, such as the Siebengebirge and Eifel.
You'll be able to get these at most decent book shops - try the Mayersche at Neumarkt, or Globetrotter just round the corner. Failing that, they're also available from amazon.de.
Once you're on the road, you can follow the white/red cycle path signs. These are pretty comprehensive and reliable, but tend to show the quietest rather than quickest routes.
